Diogo Fróis was son of Gaspar Clemente and his wife Inês Cardim. He was also brother of Jorge Cardim Fróis, inspector judge, client ombudsman e associate judge in different instances of the regal administration; of Fathers Fernão Cardim and Lourenço Cardim, both priests of the Companhia de Jesus; and of the kingdom’s Chief Surgeon, Dr. Manuel Cardim.

The existing bibliographic references about him state that he was a visiting professor of moral theology at the college and university in Coimbra when, during the plague in Lisbon (1568-1569), while assisting the inhabitants, he was contaminated and died at the city hospital.
